Effects of Temperature Pretreatment , Growth Regulators and Antibiotic Treatments on Anther Cultures of Various Cultivars of Garlic (Allium sativum L.)


Abstract
Effects of temperature pretreatment, growth regulators, and antibiotic treatments on callus formation and plant regeneration in anther cultures of various cultivars of garlic were investigated. The number of callus-formed anthers was higher on BS medium containing 2.0 §·/§¤ NAA than on other combinations of growth regulators. Pretreatment of anthers at 5¡É for 1-2 days was favorable for callus formation. However, pretreatments at high (35¡É, 80¡É) or extremely low temperatures (-70¡É) were not effective in garlic cvs. Cheju and Nagano. Among the 44 cultivars tested, none was found suitable for anther culture. In terms of callus formation and plant regeneration, no significant differences were observed between the southern type and northern type garlics, or between local and foreign cultivars. The addition of tetracycline, ampicillin, and gentamycin in dipping solution for flower stocks and in culture media had no effect on PMC development and callus formation. All regenerated plantlets were diploids (2n = 16).
